Clobetasol (Clovate Cream) belongs to a group of drugs known as glucocorticoids and is used for the short-term topical treatment of inflammatory and pruritic manifestations of corticosteroid sensitive moderate to severe dermatosis of the scalp. It is also very effective for contact dermatitis due to exposure to poison ivy/oak.
Clovate uses
These include the following:
- Psoriasis (without disseminated plaque psoriasis)
- Lichen planus
- Recalcitrant dermatoses
- Discoid lupus erythematosus
Mechanism of action:
It is believed that corticosteroids act by inducing proteins that inhibit phospholipase A2, collectively called lipocortins. It has been postulated that these proteins control the biosynthesis of potent mediators of inflammation, such as prostaglandins and leukotrienes, by inhibiting their common precursor’s release arachidonic acid. Phospholipase A2 releases arachidonic acid from membrane phospholipids. However, Clobetasol binds like other corticosteroids to the glucocorticoid receptor, which penetrates the nucleus and transcriptional genetics (transrepression/transactivation) changed.
